A Matlab-Based Approach for Estimating the Area Taken Up by Cells Attached to Micropatterned Optically Opaque Surfaces

Abstract Cell attachment is of paramount importance in implant design, bioreactor tissue engineering and the design non-fouling surfaces. Surface roughness a significant factor that affects cell attachment. To explore impact characteristics, micromachining approaches can be used to fabricate surfaces with controlled microscale topography. When optical microscopy employed study optically opaque micropatterned surfaces, one needs separate area an image coated cells from background. Manual counting assess amount attached cells. However, this process very time consuming, when studied surface larger than several square millimeters. This paper describes approach for automatic estimation micro-patterned platforms. Saccharomyces cerevisiae yeast were test developed approach. The uses registration segmentation tools available MathWorks MATLAB R2020b Image Processing Toolbox. factors affect accuracy (magnification, contrast focus) as well ways improving results are discussed.
